【摘要】 目的 探讨西格列汀对糖尿病大鼠肾脏8-羟基脱氧鸟苷(8-OHdG)和超氧化物歧化酶(SOD)表达的影响。 方法 复制糖尿病大鼠模型,随机分为西格列汀1 mg/(kg·d)干预组(Sita1,n=15),西格列汀10 mg/(kg·d)干预组(Sita2,n=15),糖尿病未干预组(DM,n=16)及正常对照组(NC,n=15)。14周末观察各组FPG、FIns、肾功能、24 h尿蛋白、8-OHdG和SOD的变化。 结果 与NC组比较,DM、Sita1和Sita2组FIns及血清SOD活性均明显下降(P<0.05),FPG、BUN、SCr、24 h尿蛋白及尿8-OHdG均明显升高 (P<0.05)。与DM组比较,Sita1组BUN、SCr、24 h尿蛋白及尿8-OHdG均明显降低(P<0.05),血清SOD活性明显升高(P<0.05);Sita2组FPG、BUN、Scr、24h尿蛋白及24h尿8-OHdG明显降低(P<0.05),FIns和血清SOD活性明显升高(P<0.05)。 结论 氧化应激参与糖尿病大鼠肾脏病变的发生发展,西格列汀对糖尿病大鼠肾脏有一定的保护作用。

【Abstract】 Objective To investigate the effect of sitagliptin on the expressions of 8-OHdG and SOD in diabetic rats kidney. Methods The rat models of diabetes were randomly divided into the following four groups: Sita1 group (Sitagliptin with the dose of 1 mg/kg body weight daily, n=15), Sita2 group (Sitagliptin with the dose of 10 mg/kg body weight daily, n=15), DM group (untreated diabetes group, n=16), and the NC group(n=15). 14 weeks later, The levels of FPG, FIns, BUN, Scr, urinary protein, 8-OHdG and SOD were detected. Results Compared with NC group, FIns and serum SOD significantly decreased in DM, Sita1 and Sita2 group (P<0.05), while FPG, BUN, Scr, urinary protein for 24 h and urinary 8-OHdG excretion significantly increased in these group (P<0.05). Compared with DM group, BUN, Scr, urinary protein for 24 h and urinary 8-OHdG excretion significantly decreased in Sita1 group (P<0.05), while serum SOD significantly increase in this group (P<0.05); FPG, BUN, Scr, urinary protein for 24 h and urinary 8-OHdG excretion significantly decreased in Sita2 group(P<0.05), in which FIns and SOD significantly increased(P<0.05). Conclusion Oxidative stress may take part in the pathogenesis of diabetic kidney injury, and Sitagliptin has some renoprotective effect on diabetic rats.
